We have HP ProDesk400 400 G2.5 SFF Desktop PC's that we are struggling to BitLocker, is this because of the age of this model, the BIOS version is 2.7 and we have upgraded to Windows 10?

That'll encrypt with BitLocker absolutely fine Dan - at least it should. It also should feature TPM too so will be fine.
Is TPM properly enabled and not hidden in the BIOS?
Even it doesn't feature TPM (which the model normally does) it should use "software" BitLocker with Win10 instead?

I comes up with an error when even trying in Windows, however it mentions about TPM in the BIOS, however I can't find it, is there a way to un-hide?

Within Windows please open a elevated command prompt and run
TPM.MSC.
This should display the known version and state of the TPM chip. If Windows can't see it at this point then it's either disabled or/and hidden in BIOS.
If Windows can't see the TPM chip BitLocker and Sophos SafeGuard will run into issues. Especially if it can see it but not control it.
Also double check in BIOS that (if possible) you enable UEFI and not legacy. Legacy will narrow your options with TPM and potentially lead to more issues.
