Thanks for reaching out to the Sophos Community Forum.
While it is possible to block removable devices based on type (Bluetooth, Secure Removable storage, Floppy, Infrared, Modem, Optical drive, Wireless, MTP/PTP), it is not possible to outright block the port on the device through Sophos Endpoint.
