I know that SQL Server 2008 R2 isn't officially supported with Enterprise Console 4.5, but does anyone know if it would anyway?

Installer and Database support for SQL 2008 R2 (Express and Server) isn't present with Enterprise Console 4.5.
You will be able to install SQL 2008 R2 and create a SOPHOS instance or upgrade SQL 2008 instance to SQL 2008 R2 instance, but you will recieve Sophos Management Service connectivity issues.
In a nutshell - I would strongly recommend waiting for Enterprise Console 4.7 which will use the SQL 2008 Express BUT contain support for SQL 2008 R2.
