Have an issue with a Mac that constantly asking for Full Disk access, I have manually moved the appropriated items to the full disk access area yet he's still receiving the prompt.

Can you tell us if you used the Sophos dialog with the drag and drop functionality, or added the items yourself? If you added them yourself, could you try steps shown in the dialog? If you have used the dialog, or that doesn't help, we'll need to look a little deeper.
In order to help us with the investigation, please provide an SDU from the affected machine by following these steps:
My Big Sur EAP users are starting to report the same issue.
I think Big Sur 11.2 is out, may have something to do with this. However, one said he is on 11.1
Bryan Wei This is a different issue, it's not related to the full disk access prompt. This was triggered by the release of macOS 11.2.
You should be able to fix it by doing the steps listed here:

Where do I upload the SDU? Never submitted it before.
Just to confirm, this issue happens for 11.1 and 11.2
Hi James, if it's run from within Central it will be automatically uploaded. We'd just need the filename from the Device page and our engineers can grab it from this end.
Hi Driss, just to confirm are you referring to the full-disk access dialog or the encryption dialog?
In either case, the version of Big Sur won't make a difference it's down to our software and the steps that have been followed.
We'd love some more information if you can.

I ran the diagnostics from the device, device name is Ryan's Macbook Pro.
Thanks James, if you ran it from the MacBook then it won't have been uploaded, instead it will be on the desktop. Please share the archive using your preferred file sharing service such as OneDrive or Dropbox. If you ran it from the device page within Central then we'll need to know the filename, it will be shown on the device page.
