Yes this is possible, for recommended approach, schedule a downtime for this activity then install individual SFP modules while the unit/s are turned off.

You would start to shutdown the slave first then Install the module but do not turn it on just yet > Then afterwards shutdown the master - this is where downtime starts. > Install the module and power on the master > Wait until all services are up and running > Check the module in Webadmin - Hardware should be up and running > You can power on the slave with the module and check the HAsync

I would also recommend you to get in touch with your local Sophos Partner/SE or if you would opt for a Professional Services for this activity and have this planned accordingly.
